Using planarian flatworms to understand organ regeneration

ScienceDaily (Oct. 25, 2012) ? In a new study published in the Oct. 16 issue of Developmental Cell, corresponding author Professor of Cell and Developmental Biology and Howard Hughes Medical Institute Investigator Phillip Newmark and colleagues report the identification of genes that control growth and regeneration of the intestine in the freshwater planarian Schmidtea mediterranea.

How animals repair their internal organs after injury is not well understood. Planarian flatworms are useful models for studying this question. After injury, they are able to re-grow missing body parts, as well as all as organs that are damaged or lost, such as brain, eyes, and intestine.

Injury initiates a complex set of cellular events. In planarians, specialized somatic stem cells called neoblasts divide and give rise to all of the different cell types required to rebuild fully functional body parts. Old tissue remaining after amputation remodels and integrates with the new cells that are produced.

The molecular signaling pathways coordinating these cellular behaviors to achieve organ regeneration have not been well characterized. David Forsthoefel, a postdoctoral researcher in Newmark's laboratory and the lead author on the study, wanted to address the problem using the planarian intestine as a "model organ," in part because so few animals are capable of repairing severe damage to their digestive systems.

"The ability to recover from loss of digestive tissue is rare in the animal kingdom," Forsthoefel said. "What we learn from how a simple worm deals with gut damage might one day help us to come up with better medical therapies, for example in the treatment of short bowel syndrome, in which segments of intestine must be removed from patients with digestive diseases, leading to impaired nutrient absorption."

Forsthoefel developed a method for purifying a single intestinal cell type from the planarian gut. He and Newmark lab members then went on to identify over a thousand genes that were uniquely expressed at higher levels in intestinal cells than in the surrounding planarian tissues. Guessing that some of these genes would have important roles during intestinal growth and regeneration, they probed the function of a subset of these genes using a technique called RNA interference, in which the expression of individual genes is selectively inhibited. They were able to pinpoint functions for specific genes, for example in the establishment of the appropriate pattern of intestinal branches, and the production of functional intestinal cells capable of taking up nutrients.

The authors also identified a transcription factor called nkx-2.2 that, although expressed in the intestine, was required for neoblasts to proliferate in various contexts, including after injury. This result suggests a potential role for the intestine in regulating stem cell division, a result Forsthoefel is following up by identifying genes downstream of Nkx-2.2 that might have more direct roles in communication between the intestine and neoblasts.

"How cells in the vicinity of damaged tissue contribute to the choices stem cells make in response to injury is an area of regeneration biology where much more research is needed," Forsthoefel said.

The field of regeneration research is rife with such uncharted territory. How do animals manage to produce the correct number specific cell types, at the correct locations? What are the signals that instruct stem cells to become specific cell types, and where do they come from? How is organ-specific morphology, for example the number of intestinal branches, determined? This study from the Newmark lab, the first systematic effort to elucidate intestinal morphogenesis in planarians, lays the groundwork for addressing many of these fundamental questions of organ regeneration.

(AP) ? Google and its street-view cameras already have taken users to narrow cobblestone alleys in Spain using a tricycle, inside the Smithsonian with a push cart and to British Columbia's snow-covered slopes by snowmobile.

The search giant now has brought its all-seeing eyes ? mounted for the first time on a backpack ? down into the Grand Canyon, showcasing the attraction's most popular hiking trails on the South Rim and other walkways.

It's the latest evolution in mapping technology for the Mountain View, Calif., company, which has used a rosette of cameras to photograph thousands of cities and towns in dozens of countries for its Street View feature. With a click of the mouse, Internet users are transported virtually for a 360-degree view of locales they may have read about only in tourist books and seen in flat, 2-D images.

"Any of these sort of iconic, cultural, historical locations that are not accessible by road is where we want to go," said Ryan Falor, product manager at Google.

Google announced the trekker earlier this year but made its first official collection of data this week at the Grand Canyon.

The backpacks aren't ready for volunteer use, but Google has said it wants to deploy them at national forests, to the narrow streets of Venice, Mount Everest and to ancient ruins and castles.

The move to capture the Grand Canyon comes after Apple chose to drop Google Maps from its mobile operating systems and opted to use its own mapping program that was derided for, among other things, poor directions and missing towns.

Steve Silverman, operations manager for Google didn't directly address the competition in saying: "Just trying to document a trail, it's going to be hard to beat this."

Google launched its Street View feature in 2007 and has expanded from five U.S. cities to more than 3,000 in 43 countries. Google teams and volunteers have covered more than 5 million miles with the Street View vehicles on a scale that other companies haven't approached, said Mike Dobson, president of Telemapics, a company that monitors mapping efforts.

"You could safely say that it's a standout, well-used application and they don't really have any competition," he said.

As the sun rose Monday, Luc Vincent, Google engineering director, strapped on one of the 40-pound backpacks and set down the Bright Angel Trail to the Colorado River ? a nearly 10-mile hike that goes from 6,900 feet in elevation to 2,400 feet. He hiked back up from Phantom Ranch, which can be 30 degrees warmer than at the rim, through the South Kaibab Trail and also gathered data on other trails.

The so-called trekker captures images every 2.5 seconds with 15 cameras that are 5 megapixels each from the rest areas, the steep switchbacks, the change from juniper trees to scrub brush and the traffic that moves aside as a courtesy to mule riders.

The GPS data is limited, so Google must compensate with sensors that record temperature, vibrations and the orientation of the device as it changes, before it stiches the images together and makes them available to users in a few months, Falor said.

Hikers that were on the trail when the data was gathered will have their faces blurred ? an attempt by Google to ensure privacy. Street View has run into problems in places like Europe and Australia for scooping up information transmitted over unsecured wireless networks.

A removable hard drive on the trekker stores the data gathered at the Grand Canyon. Tourists looked at the trekker strangely this week, as if it was something from outer space.

Sharon Kerfoot, a first-time visitor from Alberta said being able to view the terrain ahead of time, gauge the difficulty of the hike and know just how wide the path is would benefit those considering a trip to the Grand Canyon. She and a group of friends headed down the same path as Vincent but on mules, not foot.

"I think it's an excellent idea to give people a broader perspective on what they're getting into," she said.

What the images won't tell visitors is how much water they should carry down the trails, how to prepare for temperature changes, what type of food to take and how much, and how best to protect the natural resources, park spokeswoman Maureen Oltrogge said.

"Stitched together with other information out there, the technology could be valuable," she said.

Markets subdued as focus remains on US

LONDON (AP) ? Financial markets were on the defensive Tuesday ahead of another round of U.S. corporate earnings, including from Facebook, and amid uncertainty over the outcome of the U.S. presidential election.

The U.S. has been center stage this week as the earnings season continues and as the election looms. So far earnings have been mixed, particularly from the technology sector.

"We are currently in the third week of a disappointing corporate earnings season," said Craig Erlam, market analyst at Alpari.

Facebook's earnings results after the market close will be key for its future. The social network has had a dire time since its stock flotation earlier this year, losing over a third of its value, as investors have grown concerned about the company's ability to raise money from advertising.

"The results have the potential to make or break Facebook as an investment," said Erlam.

Over-arching the focus on earnings is the battle for the White House, with opinion polls showing the two candidates are in a dead heat. Last night's debate between President Barack Obama and Mitt Romney appears to have done little to change that.

Meanwhile, the Federal Reserve starts its regular two-day policy meeting Tuesday but little is expected in the midst of the election.

By late morning in Europe, Germany's DAX was down 0.9 percent at 7,265 while the CAC-40 in France fell 0.6 percent to 3,464. The FTSE 100 index of leading British shares was down 0.8 percent at 5,837.

Wall Street was poised for a lower opening, with Dow futures down 0.5 percent and the broader S&P 500 futures 0.7 percent lower.

In Europe, there was little scheduled news regarding the debt crisis to drive investor sentiment. Over the past few weeks, markets have been shored up by hopes that Spain will soon tap a new bond-buying facility from the European Central Bank and expectations of further stimulus measures from Beijing.

"With the main market drivers being a Spanish bailout request and substantial Chinese stimulus being largely priced in and having not come to pass, investors are growing increasingly nervous," said Mike McCudden, head of derivatives at Interactive Investor. "Furthermore, without another EU Heads of State meeting until mid-December there is growing concern that we will not see any progress in the eurozone until then."

Earlier in Asia, stock indexes fared slightly better. Japan's Nikkei 225 index eked out a marginal gain to close at 9,014.25 a day after the country's currency fell to a three-month low against the dollar.

A weak yen helps Japan's mammoth export sector by raising the value of company profits repatriated from abroad. Still, grim export data ? which Monday showed a 10 percent drop in exports for September compared with a year earlier ? kept advances in check.

South Korea's Kospi declined 0.8 percent to 1,926.81 but mainland Chinese stocks lost ground, with the Shanghai Composite Index down 0.9 percent to 2,114.45. The Shenzhen Composite Index lost 1.4 percent to 869.96.

Hong Kong markets were closed for a public holiday.

In the currency markets, the euro was down 0.3 percent at $1.3033 ? the euro often suffers when investor appetite for taking on risk is diminished.

As do oil prices ? benchmark oil for December delivery was down 14 cents to $88.51 per barrel in electronic trading on the New York Mercantile Exchange.

Election's other crystal balls: Football, baseball, basketball

Why invest in polling and television ads, when rooting for a few oddly dispersed teams can lead to presidential victory?

Election watchers have been looking for correlations between sports and the presidency for years. A few have been found, explaining why challengers should root for the Washington Redskins, Democrats should root for the National League baseball teams and Republicans should cheer for the Los Angeles Lakers.

The most well known sports related presidential predictor is the Redskin's Rule. The rule is simple: if the Redskins lose, the challenger wins; if the Redskins win, the incumbent wins. Since 1940 (the 'skins moved to Washington in 1937), the outcome of the Redskins last home game before the election has correctly predicted the outcome of the presidential election 17 out of 18 times.

The Redskins play the Carolina Panthers at home on November 4, just two days before the election.

If you believe the rule, Obama will be cheering for the Redskins and Romney will be cheering for the Panthers.

Major League Baseball, in a press release, noted another correlation from the twenty-six World Series played since 1908, albeit a less reliable one.

About three-fifths (59%) of the time that the American League team won the World Series, the Republican candidate won the White House. Likewise, 67% of the time that the National League won the World Series, the Democrat won the White House.

After a long season, the National League's San Francisco Giants will face off against the American League's Detroit Tigers in the World Series starting on Wednesday. If the odds hold, Romney - a native of Michigan - will be pulling for the team from Detroit, while Obama will be pulling for the Giants.

Though not totally accurate, this theory has correctly predicted the last three elections.

But that is not all - the United States is more sports and politics obsessed than just two theories.

GOP's McMahon ad targets Conn. Obama voters

GOP Senate candidate Linda McMahon is courting ticket-splitters in a new ad in which four supporters of President Barack Obama, who has a double-digit lead in the state, say they also will cast ballots for her. Her campaign strongly denied she was writing off Republican presidential hopeful Mitt Romney, saying she supports the former Massachusetts governor "100 percent."

"Linda McMahon will work with President Obama and I believe he'll work with her," says one of the voters in the ad, David Cole, a disabled veteran who said he plans to vote for the Republican on the independent line in November. McMahon's name will appear as both the Republican and the Independent candidate.

Nonsense, said Rep. Chris Murphy, the Democrat in the race once thought to have a better shot at winning.

"This is one of the strangest political ads I have ever seen because we know that Linda McMahon is going to oppose President Obama on everything that he stands for. She's told us that, over and over again," said Murphy, referring to the Republican candidate's calls to repeal and replace Obama's health care legislation and her opposition to his positions on energy reform and tax policy. "I mean, there is almost nothing Linda McMahon agrees with President Obama on, which makes this ad absolutely deceitful."

It was the latest volley in one of the nation's quirkiest Senate races, one of a series of stubbornly tied contests that have left the national battle for control of the Senate in question. Republicans need to gain three seats, or four if Obama wins. From Massachusetts to Connecticut, Virginia, Indiana, Missouri, Wisconsin, North Dakota, Montana, Nevada and Arizona, contests for Senate are so tight that with two weeks to go, the parties, candidates and outside groups are doling out cash and getting creative about peeling votes away from the other guy.

With McMahon's ad, Connecticut joins Senate races in which candidates are banking on voters choosing candidates of different parties down the ticket. In places like Montana and North Dakota, the politics are reversed, with Romney expected to win big and Democratic Senate hopefuls making their pitches as independent-minded candidates.

In Connecticut, the candidates are racing to win the seat of a retiring senator who counted himself an independent: Sen. Joe Lieberman, who caucused with the Democrats.

The state has a vibrant independent streak. As of last week, the Secretary of the State's Office reported 2 million registered voters in Connecticut, with the largest group, 842,335, being unaffiliated. They are followed by registered Democrats, at 744,729 and registered Republicans at 422,312.

McMahon's ad rankled Republicans, after they chose the former wrestling empire executive over former Rep. Christopher Shays in the August primary. McMahon lost in 2010 to then-Attorney General Richard Blumenthal after spending more than $50 million of her own money. She has so far personally spent some $27 million on this year's race. State GOP Chairman Jerry Labriola said the Connecticut Republican Party headquarters has been "experiencing very high call volume in response to the ad," with Republican "expressing serious concerns."

"It's a distressing situation, but we're doing our best to deal with it," he said, adding how he still believes Romney can win in Connecticut and how he's "working very hard to get to elect all Republicans on the top line of the ballot this November."

McMahon's campaign spokesman, Todd Abrajano, said the campaign has not received a large number of complaints.

"I think most people realize that in the state of Connecticut, if you're a Republican running statewide, you're going to have to get independents and Democrats to vote for you in order to win," said Abrajano.

Abrajano said McMahon supports the GOP presidential ticket "100 percent" and is not encouraging people to vote for Obama, who is expected to win relatively easily in Connecticut. However, if Obama wins, he said McMahon is willing to work with him.

Bill Heyn, a 70-year-old retiree from New Canaan who attended a McMahon rally on Monday in Stamford featuring New Jersey Gov. Chris Christie, said he understands why McMahon is running such an ad.

"It makes sense. She's got to get somebody to cross over," he said. "She's trying to win."

Simply stretch the fabric or paper over the outside perimeter of the inner ring and trim to size. Screw on the outer ring and hang on a simple nail or picture hook.

Cabinet Hardware: Make use of extra cabinet hardware such as small knobs or cup pulls to create a useful and stylish jewelry organizer. Start by wrapping a small piece of luan or plywood board in your fabric of choice. Small knobs can be used to hang necklaces or bracelets from, French wire earrings can be hung on small drawer rods and inverted cup pulls (bottom side up) can hold small items such as brooches and rings. Determine the placement of the hardware and screw into the wood. Mount the organizer right on the wall. I'll have a posting order made before we get things rolling, we may not get up to 11 or more though so no real reason to worry about that yet.

As for balance, this game will be strictly RP, and doesn't have to adhere to the rules quite as much as if we brought out the books and dice. If I find a character that I believe is OP or broken in any way I'll whip out the nerf-stick and beat it until everyone's happy.

The classes, every base class is up for grabs IF you can make it believable. Minotaur psions and pixie samurai will need an excellent backstory before I say yes. As for prestige, and multiclassing, as we progress through the storyline PM me about it and I'll try to squeeze in some NPC trainers... I'm pretty much open to everything at this point, there's a lot more freedom to be had since it's going to be strictly roleplay.

All that I ask is that the characters be believable.

In Slate?s Homeland TV Club, June Thomas will IM each week with a different partner?policy experts, intelligence researchers, critics, and even Slate commenters. This week she chats with Slate copy editor Miriam Krule.

June Thomas: Miriam, we've got to begin with the I-didn't-see-that-coming final scene. Homeland has a long history of great cliffhangers, but seeing Brody being led away in handcuffs and with a hood on his head leaves me with no idea of what's coming next?and we're only one-third of the way into the season. What do you think of this plot twist?

Miriam Krule: I definitely had that reaction at first, too. I was getting excited for some Wire-style surveillance, but then I remembered that on The Wire they were trying to catch the guy; here they already had him. There wasn?t much about Brody that we viewers didn?t know, so how much surveillance-related drama could there possibly be? In Season 1, it was all about what we didn?t know?Season 2 has to deal with Brody?s true nature so there?s no real suspense in watching him do his thing for an extended period of time. The one thing I will miss about the surveillance room is the strange Carrie-Quinn dynamic.

Thomas: Yes, I feel the same way. It's been a while since we met a new character, so I was looking forward to the Carrie-Quinn sparring?and to hanging out with Virgil and Max. It almost seemed like Carrie was going to investigate Quinn the way she investigated Brody. And I suppose that's not a crazy thought?after all, this season she seems to be really going with her instincts, whether that involves ignoring everyone's instructions not to return to Abbas' apartment in Beirut or blowing up the surveillance by going up to Brody's room.

Why do you think she did it? She said she "saw ... in his eyes" that he was onto her. Do you believe that? I almost thought she was mad at him for bringing up her ECT.

Krule: I thought the scene at the CIA, where she ?bumped into? Brody, was Carrie at her best?she did the most amazing, awkward encounter where she was completely in control. In their second interaction, he had all the power (he was the one that called, after all), and she was just reacting, something we?ve seen she?s not very good at. So her reaction was to try and take the power back. I don?t know what she ?saw ? in his eyes,? but once he called, I had a feeling she would find a way to crack.

Can we back up a second, though? I?m confused about Carrie?s reinstatement. Yes, she was right about Brody, but that?s not why she was kicked out of the CIA. It seems crazy to me that David Estes would a) apologize b) let her back in. Wasn?t the issue that she?s bipolar? Shouldn?t they have realized that she would do something like this?

Thomas: I didn't think she was back in the CIA, but rather that Estes was using her as a non-company asset (same as Virgil and Max) for an off-the-books investigation. It's off-campus, no one at the CIA knows about it ... but Estes was responsible for renting that lovely bit of real estate and for sending over a pile of sweet CIA surveillance equipment and Quinn to "run it."

Not to beat a dead horse with the credibility factor, but ... I didn't buy that. Something that was bound to be as big a deal as a former prisoner of war/current congressman being revealed to be an al-Qaida sleeper couldn't remain a secret. And even if my long-simmering suspicions about Estes were true, and he was sneakily sabotaging the whole thing, why would Saul Berenson be sitting around the room like a broken toy just waiting for something to happen? He goes from being the Beirut station chief to a guy hanging out at an off-the-books operation?

Krule: I was about to bring that up! What happened to Saul? Yes he?s had a lot happen to him?his wife and prot?g? both left him in different ways last season?but after going door-to-door with the Brody video (that was strange and against protocol, surely)?he seems content to just mull about in the background. The more Mandy Patinkin, the better is my philosophy. My only, slight, consolation is Dana, who?s been filling the Sally Draper-sized hole in my heart. I?m usually not a fan of the family side plots, but I?m a sucker for Dana.

Thomas: You're right that there was a ton of interesting family stuff this week, and it began in the very first scene with Kenny Estes (the youngest person in the show) in Darth Vader garb telling Saul (maybe the oldest): "I am your father. Don't make me destroy you." Cue lots of family destruction: Nick Brody moving out of the family home, Estes cheating on the agency, and Saul acting like the impatient dad watching Carrie and Quinn bickering. But we also saw a little bit of bonding between Dana?who, I agree, always brings something to the table?and Finn. They're the next generation, complete with Dana dissing the old, dumb vice president. I have to say, though, kids today have some kinky text habits--she's Sally Hemings to his Thomas Jefferson? That's up there with the uncomfortable racial politics of the surveillance team focusing on "the dark-skinned" people that Brody interacted with. It must?ve been icky for Estes?the only person of color in the room at the time?to hear that, even as he accepts that it made sense. And how strange that Roya Hammad somehow became an honorary white woman. The power of the press!

Krule: One thing we haven?t talked about that?s been bubbling up for a while is how Brody?s former Marine friends have essentially solved what the CIA couldn?t and are the only ones asking any questions. Lauder bangs on Brody?s door (in a manner somewhat reminiscent of Carrie running on the lawn in last season?s finale) and basically demands to talk to Brody/find out what really happened to Tom Walker?only to be talked down by Mike. I couldn?t shake the feeling I had during Season 1?and it?s one of my biggest frustrations with the show?that Homeland seems obsessed with having ?crazy people? be right about everything, and all the ?sane/rational? people preventing them from doing what needs to be done.

Thomas: I know you're a big fan of Hatufim, the Israeli show that Homeland is based on. How would you compare the two shows?

Krule: Yes! I started watching Hatufim when Hulu began streaming it (as Prisoners of War) this summer, when I was anxiously awaiting Season 2 of Homeland. Although the American show is loosely based on the Israeli one, and many of the plot points overlap, where Homeland is full of suspense, Hatufim is full of emotion, for lack of a better word, making them very different shows. This makes sense: After all, Israelis are just celebrating the one-year anniversary of Gilad Shalit?s release, and they have an appetite for storylines about POWs coming home. The U.S. audience needs the added elements of suspense and intrigue. For example, the Carrie Mathison character is not only a minor presence on Hatufim, she is told to pose as one of the POWs? girlfriends (there are two in the Israeli show). Most of the focus is on the families and the internal drama. As a result, the pace of Hatufim is slower, and it focuses more on adapting and is therefore more true to life. When I watch Homeland, I?m usually at the edge of my seat. When I watch Hatufim, I feel emotionally drained?in the best possible way. Oh, and the Israelis were POWs for 17 years, more than double Brody?s absence.

Going back to your first question about what?s going to happen next, I?m most curious to see what they tell family and friends about where Brody is. I can?t imagine they?ll make some big announcement and let the public know. Then the show really would be over. My best guess is that they will try to turn him against Abu Nazir. The Carrie-Brody dynamic has to continue in some capacity, and this seems like the best bet.

Are Democrats finding ways around campaign finance laws?

Suppose that Obama loses the election. ?He obviously isn't going to run for any other office in the future. ?So if his campaign gets a loan, it would seem fairly likely that the campaign would default on the loan. ?The Free Beacon has this interesting information:
Obama For America took out a $15 million loan from Bank of America last month,?according to the campaign?s October monthly FEC report. The loan was incurred on September 4 and is due November 14, eight days after the election. OFA received an interest rate of 2.5% plus the current Libor rate.?
Warren Buffett, Obama donor and namesake of the infamous ?Buffett Rule,??invested $5 billion?in Bank of America last year in an effort to help the ailing financial institution. . . .
Surely this is a fairly high risk loan and it doesn't seem like the interest rate on this loan comes anywhere near to close to covering that risk. To judge whether this loan is fair, it would be interesting to know what type of collateral has been offered to secure it. ?This certainly seems like one way to get around the campaign finance rules.

Contrast this loan with a loan to the Democratic National Committee:
The Democratic National Committee (DNC) now owes $15 million to the union-owned Amalgamated Bank of New York,campaign finance records show.
The DNC received a $7 million loan from the bank in September, in addition to the?$8 million loan?it took out the previous month, neither of which has been paid back. The loans account for the majority of the committee?s $20 million in total debt.
The DNC paid Amalgamated Bank more than $18,000 in loan interest for the month of September, the records show.
Amalgamated Bank, often described as ?America?s Labor Bank,? is a national entity, the majority of which is owned by the Service Employees International Union (SEIU), a large, politically active union with deep ties to the Democratic Party. . . .

The big difference is that the DNC will be around after the election. ?Though even in that case, it would be useful to make sure that the entire loan is eventually paid off.

(AP) ? Israeli soldiers commandeered a vessel carrying pro-Palestinian activists destined for Gaza on Saturday, cutting off communications and steering it from international waters toward the Jewish state.

The ship was the latest in a series of activist-manned vessels challenging Israel's blockade on the territory, imposed when the militant group Hamas seized the coastal strip in 2007. Israeli officials say they need to maintain the blockade, mainly to prevent weapons smuggling.

Six Israeli naval vessels stopped the Estelle as it was about 30 nautical miles from Gaza, with masked soldiers boarding the ship and ordering it to sail to Israel's Ashdod port, said a Victoria Strand, a spokeswoman for the activists.

The Swedish-owned, Finnish-flagged ship left Naples, Italy, on Oct. 7 with about 30 people from eight countries on board, including lawmakers from Norway, Sweden, Greece and Spain, as well as Israeli activists and a 79-year-old former legislator from Canada.

Israel, aided by Egypt, imposed a full border closure of Gaza by air, land and sea after Hamas took over the strip and drove out forces loyal to Palestinian President Mahmoud Abbas. Israel eased its restrictions after its raid of a Turkish-led blockade-busting flotilla in 2010 left nine activists dead and sparked international condemnation.

Still, Israel continues to block sea access to Gaza and severely restricts its ability to export goods and import raw materials.

Israeli military spokeswoman Lt. Avital Leibovich accused the activists of staging a provocation and said the naval blockade was necessary to safeguard Israel's security.

"We have this blockade because there are constant smuggling attempts of weapons, munitions that eventually reach the hands of terror organizations inside Gaza," she said.

Strand said the takeover of the Estelle by Israeli forces was a "demonstration of ruthlessness."

The ship carried cement, basketballs and musical instruments. It was emblazoned with "Ship to Gaza" on one side, and also flew the colorful red, green, black and white Palestinian flag.

Activists say the blockade amounts to collective punishment of Gaza's 1.6 million residents, denying them the chance to trade and travel freely. Neighboring Egypt continues to impose restrictions at its passenger crossing with Gaza. The blockade has deepened the hardships of Gaza's most vulnerable in the impoverished territory, where between 70 percent to 80 percent of all residents rely on U.N. donated food aid to get by.

"It's hard to imagine what threat one sailboat, loaded with humanitarian supplies and a small number of people, could do to" Israel's mighty military, said Eva Manly, the wife of former Canadian parliamentarian James Manly. She said she lost contact with her 79-year-old husband early Saturday.

Israeli Defense Ministry spokesman Joshua Hantman said the goods onboard would be checked before entering Gaza through an Israeli-controlled land crossing. Hantman said militants have attempted in the past to smuggle weapons by sea. The latest attempt was in 2011, when a vessel carrying 50 tons of weaponry sought to reach Gaza, he added.

Fasting hormone helps mice live longer

New work suggests that a hormone that makes the body think it?s starving could prolong life about as long as severely cutting calories does but without the denial.

A hormone called fibroblast growth factor-21, or FGF21, lengthened the lives of mice that had been genetically engineered to constantly produce large amounts of the protein, researchers at the University of Texas Southwestern Medical Center at Dallas report online October 15 in eLife. The hormone is normally made by the liver during fasting and may tap into some of the same life-extending biochemical processes as does caloric restriction, a proven longevity booster.

Caloric restriction ? usually defined as cutting calorie intake to 75 to 80 percent of the amount needed to maintain normal body weight, while still maintaining good nutrition ? has been shown lengthen life in a wide variety of species, such as fruit flies and dogs. Minimal calorie consumption turns on many different biological processes that slow aging, says Cynthia Kenyon, a developmental biologist at the University of California, San Francisco. The hormone in the study somehow interferes with a chain reaction anchored by insulin-like growth factor-1 (IGF-1), a process that is also shut down by caloric restriction and thought to be responsible for many of its life-extending effects.?

In the study, researchers led by UT Southwestern?s David Mangelsdorf and Steven Kliewer genetically engineered mice to constantly make five to 10 times as much FGF21 as normal. These engineered mice lived 30 to 40 percent longer than normal mice on a standard diet. Female mice benefitted from the hormone even more than males; about a third of the FGF21-producing female mice still were alive at 44 months old. Average survival for normal mice in the study was about 28 months.

Hormone-producing mice were leaner than normal mice despite eating slightly more. They also had improved health measures, such as greater sensitivity to insulin and lower triglyceride levels. The mice tended to burned fat instead of sugar for energy, the researchers found. But the news was not all good. Higher levels of FGF21 were associated with low bone density and female infertility. Both drawbacks are part of the body?s normal response to starvation and could pose problems if the hormone is used to treat people, says Mangelsdorf, a Howard Hughes Medical Institute investigator. The researchers have already begun testing whether the hormone can extend longevity in mice if its production is delayed until adulthood.

Exactly how the hormone works is still under investigation, Mangelsdorf says. The researchers know FGF21 is made in the liver and indirectly affects that organ?s function, but they aren?t sure where else in the body it works. It is also not clear how the hormone prolongs life. It does not affect the same biological processes as caloric restriction, except for the IGF-1 pathway in the liver, yet has many of the same benefits.

But the hormone must be doing something more than simply shutting down IGF-1, says Luigi Fontana, a physician and gerontologist at Washington University in St. Louis. People who have mutations that inactivate IGF-1 are dwarves who generally don?t get cancer, but they don?t live much longer than anyone else, so the longevity benefit of the hormone may be due to some other action. Anti-aging properties of the hormone probably depend upon multiple different effects, and turning off only one biological process won?t work, Fontana says. ?Many biologists are looking for the magic bullet,? he says, ?but maybe there isn?t one.?

Endeavour's 'Shuttle Xing' Signs on Sale as 'Mission 26' Memento

It is hard to upstage something as large and unusual as a space shuttle moving through the streets of a major city, but an orange and black road sign that signaled the shuttle's crossing almost did.

Now, that scene-stealing, diamond-shaped "Shuttle Xing" sign can be yours, direct from the traffic control company behind its creation.

As the shuttle Endeavour rolled for three days through the streets of L.A., from Los Angeles International Airport to the California Science Center, Oct. 12-14, the millions who turned out to watch the museum-bound orbiter pass by photographed the winged spacecraft from every angle.

Many also took notice ? and snapshots ? of the 4 by 4 foot (1.2 by 1.2 meters) custom aluminum road signs that were placed along the space shuttle's route. The reflective orange markers featured a black silhouette of the orbiter in profile and the words "Shuttle Xing," or "Shuttle Crossing."

"We love this road signage!" the California Science Center (CSC) wrote on Twitter, in response to a photo of one of the signs that was taken by collectSPACE.com outside of the airport.

NASA also featured the sign on its website, in an "Image of the Day" titled "Shuttle Endeavour Crossing." [Photos: Shuttle Endeavour on L.A. Streets]

The same "Shuttle Xing" traffic sign was present outside the landmark Randy's Donuts where Toyota was filming its Tundra pickup truck towing the Endeavour across the 405 freeway, and on news sites such as CNN.com.

The sudden popularity of the sign inspired miniature and full-size knock-offs to be offered on eBay, fabricated in aluminum and in plastic. Replicas of the simple but iconic design quickly showed up for sale on buttons and bumper stickers.

Now the signs' original creator, Traffic Management, Inc., has begun selling the "official traffic sign" for the science center's "Mission 26: The Big Endeavour" parade through its traffic products online store.

The souvenir signs, all produced out of 0.063 inch (0.16 centimeter) aluminum, range in sizes from a foot (30.48 cm) to the full-size 48 inches (1.2 meters). Prices for the six different sizes of signs offered range from $25 to $175.

As for Endeavour, it won't be needing any further crossing signs, at least not for the next several years.

Offloaded from its four self-propelled, computer-controlled transporters that delivered the shuttle for display to the California Science Center, the orbiter is now housed inside the new Samuel Oschin Space Shuttle Endeavour Display Pavilion. The exhibition hangar opens to the public on Oct. 30.

Endeavour's next and final move won't come for another five years, when the CSC plans to have ready the space shuttle's permanent exhibit inside the still-planned Samuel Air and Space Center, opening in 2017.
