STI Rate Higher Among Military Women Than Civilian Women ...

The rate of STIs among military women is higher than that in civilian women, according to a recent study in the Journal of Women's Health.

The study, conducted by researchers at Women & Infants Hospital of Rhode Island, shows that the STI rate is seven times higher among military women than civilian women.

The researchers also found that only about one in three unmarried military women who are sexually active used a condom during the last time they had sexual intercourse, and that nearly two in three of them had more than one sexual partner over the last year.

The findings are based on reviewing past research on STI rates among military members.

"These high-risk sexual practices likely contribute to chlamydia infection rates that are higher than the rates in the U.S. general population," the researchers wrote in the study.

"To address the reproductive health needs of military women, investigations into the prevalence of unsafe sexual behaviors and consequent infection among older servicewomen and women veterans are needed," they wrote. "Direct comparison of military and civilian women is needed to determine if servicewomen are a truly high-risk group."

According to the latest data on STIs in the United States, there were about 1.4 million cases of chlamydia, nearly 322,000 casse of gonorrhea, and 13,970 non-congenital cases of syphilis reported in the U.S. in 2011. Young people and gay and bisexual men are at the highest risk for STIs, according to the Centers for Disease Control and Prevention.

analysis projects that support programs and processes from across all areas of the College. The position works collaboratively with staff at

The Benefits/Financial Analyst researches and analyzes renewal data, evaluates plan design changes including impact to budget, and make recommendations for change. The position is responsible for ensuring reporting/compliance requirements are met under relevant Federal and State law, including the Employee Retirement Income Security Act (ERISA), Consolidated Omnibus Budget Reconciliation Act (COBRA) and the Health Insurance Portability and Accountability Act (HIPAA). The Analyst is responsible for developing and monitoring the annual benefits budget, reconciling the monthly billing process and managing day-to-day benefits administration that includes providing consultation and education to faculty/staff. The Benefits Manager serves as the primary point of contact for all benefit inquiries and handles the day-to-day benefits administration for all of the College's benefits plans. The Benefits/Financial Analyst reports to the Associate Vice President for Finance.

Qualifications should include a bachelor's degree in accounting, business administration, human resources or related field and and relevant experience; benefits administration and/or compliance/audit experience preferred. Candidates should possess demonstrated experience in data analysis and report development. The position requires strong knowledge of relevant Federal and State laws, compliance issues and annual reporting under ERISA. The position requires the ability to work proactively and communicate clearly with a wide range of audiences; exceptional verbal and written communication skills are essential as is the ability to manage multiple projects simultaneously. Strong organizational and analytical skills are required; candidates must possess demonstrated ability to analyze and synthesize large volumes of data into useable formats. The position requires a strong technology orientation with experience using Microsoft Office Suite (advanced knowledge of Excel), Gmail, HRIS and/or benefits administration systems, and Internet research. Candidates should have the ability to work independently, be creative in the approach to work and possess keen attention to detail.

LeBron James at the WH: 'Mama, I Made It!'

WASHINGTON - It's LeBron James' world and we're all just living in it, according to President Obama, who honored the NBA champion Miami Heat today at the White House.

"I am honored to welcome a little up-and-coming basketball team to the White House," Obama joked as he stood with the players before a packed audience in the East Room of the White House.

"This team traveled a long road to get to where they are. In 2011, the Heat got all the way to the finals only to come up short. But when you fall, the real test is whether you can ignore the naysayers, pick yourself up and come back stronger. And that's true in basketball, but it's also true in life. That's exactly what these guys did," he said.

Obama singled out several of the team's star players, including James.

"We saw him pull off one of the greatest performances in basketball playoff history," he said of his performance during Game Six of the Eastern Conference Finals. James scored 45 points, grabbed 15 rebounds and dished out five assists to lead the Heat to a 98-79 victory in a game in which they were facing elimination.

Calling Dwyane Wade the "heart and soul of this team," Obama said he was committed to doing whatever it took to win.

"And at the post-game press conferences, he dressed well enough to land himself in GQ magazine," he said to laughter. "Show them your kicks here, Dwyane."

At one point, James interjected and asked if he could say something.

"You can if you want. It's your world, man," Obama said.

"On behalf of myself and my teammates, we just want to thank you ? for the hospitality, for allowing us to be, I mean, in the White House. I mean, we're in the White House," James said with excitement. "We're kids from Chicago and Dallas, Texas and Michigan and Ohio and South Dakota and Miami. And we're in the White House right now.

"Hey, Mama, I made it!" he exclaimed.

No sooner was support for 128GB NAND flash storage capacities found in iOS 6 beta 5, than reports pop up saying Apple is readying 128GB "ultimate" versions of the iPad 4 for launch. The news come by way of Mark Gurman of 9to5Mac, who typically has excellent sources on imminent product releases.

We?ve gotten word from sources that Apple is preparing to release (in some fashion) a single, additional iPad model. This would be a new SKU for the current fourth-generation iPad with Retina display line. At this point, we do not have pricing information, so it is definitely difficult to pinpoint exactly what this new iPad model is.

The code names are listed as P101 Ultimate and P103 Ultimate. Gurman stresses that it's not a new iPad, or iPad 5, or anything like that, simply a new, higher capacity version of the current iPad 4, in both Wi-Fi and Wi-Fi + Cellular, and in both white and black.

In early 2008, Apple bumped the original iPhone from 8 to 16GB, so this type of new SKU launch isn't unprecedented. While it lacks the oomph of a completely new model, given that the iPad was refreshed in both March and October of 2012, it does at least give Apple something new on the market, and at 128GB, and presumably at the pricier end of the line-up, at the top of the market.

No word yet on 128GB versions of the iPad mini, or of the iPhone 5 or iPod touch 5.

A family's dream home rises from the one they lost - KTIV 4 Sioux City

NORTH SIOUX CITY, SD (KTIV) -

There are those who say they have no reason to come back to Riv-R-Land Estates.? But one family says their community ties are stronger than mother nature's will.

And while floods never come at a good time for anyone, the Lenz family had just brought home a new baby in 2011 when flood waters started to rise.

"We'd seen it high before, didn't really believe it until it started happening," Todd Lenz said.

The Missouri River flooded their home, and damaged it beyond repair. They were devastated.

"What's gonna happen next? What do we do?" the Lenzs wondered.?

That's when members of the Redeemer Lutheran Church in Sioux City intervened.?

"The Lenz family out of our 1600 members are the only ones who lost their homes totally from the flood," Pastor David Zirpel from Redeemer Lutheran said.

Redeemer Lutheran Church members are used to helping out in times of disaster like hurricanes and?tornadoes. But the 2011 flood kept them close to home.

"So when the flood came up and one of our families lost their homes totally to the flood we decided to stay here and help them," Pastor?Zirpel said.?

"It was just unbelievable, just a snowball effect. Things started happening, and contacts were being made, and people were coming forward and all these volunteers were here," Jaime Lenz said.

Over ten months their church worked to rebuild their home on the spot their old one once stood. Sunday they celebrated with a special blessing, and with those who helped. They say the future now looks a lot different.

"It looks pretty promising, we're very blessed to meet all of our Redeemer friends and family and everyone involved in the project,"

The Lenz say they cannot forget this experience and say they plan to pay it forward by volunteering in the future.

Poultry producers can reduce bacterial cross-contamination in poultry cages by treating the cages with forced air that's been heated to 122 degrees Fahrenheit, according to a study by U.S. Department of Agriculture (USDA) scientists.

While being transported in coops on trucks, poultry that have bacteria such as Campylobacter can contaminate, through their feces, other poultry that are free of pathogens. Those disease-causing bacteria can then be passed on to the next group of birds during the next trip, and so forth, unless the cycle is broken.

Campylobacter is a food-borne pathogen that can be present in raw or undercooked poultry. Since the bacteria are commonly found in the digestive tracts of poultry, they're readily deposited onto coops and trucks when contaminated animals are transported to processing plants.

In the study, Agricultural Research Service (ARS) microbiologists Mark Berrang and Richard Meinersmann collaborated with researcher Charles Hofacre of the University of Georgia at Athens. Berrang and Meinersmann work in the ARS Bacterial Epidemiology and Antimicrobial Resistance Research Unit in Athens. ARS is USDA's principal intramural scientific research agency, and this research supports the USDA priority of promoting food safety.

The researchers tested the use of hot flowing air to speed the process of drying soiled or washed cages to lower or eliminate detectable Campylobacter on cage flooring.

When the hot flowing air was applied to fecally soiled transport cage flooring samples for 15 minutes after a water-spray wash treatment, Campylobacter levels declined to an undetectable level. Static heat at similar temperatures was not nearly as effective, and unheated flowing air was moderately effective, but less so than hot flowing air.

Washington casts wary eye at Muslim Brotherhood

WASHINGTON (AP) ? President Barack Obama begins his second term straining to maintain a good relationship with Egypt, an important U.S. ally.

That country's president is a conservative Islamist walking a fine line between acting as a moderate peace broker and keeping his Muslim Brotherhood party happy with anti-American rhetoric.

The White House last summer had hoped to smooth over some of the traditional tensions between Washington and the Brotherhood, when Egypt overthrew dictator Hosni Mubarak (HOHS'-nee moo-BAH'-rahk) and picked Mohammed Morsi as its first democratically elected leader.

But a spate of recent steps ? from Brotherhood-led attacks on protesters to revelations of old comments by Morsi referring to Jews as "bloodsuckers" and "pigs" ? have raised alarm among senior U.S. officials and threatens $1 billion in American aid to Egypt.

'Barrier of bodies' trapped Brazil fire victims

SANTA MARIA, Brazil (AP) ? A fast-moving fire roared through a crowded, windowless nightclub in southern Brazil early Sunday, filling the air in seconds with flames and a thick, toxic smoke that killed more than 230 panicked partygoers, many of whom were caught in a stampede to escape.

Inspectors believe the blaze began when a band's small pyrotechnics show ignited foam sound insulating material on the ceiling, releasing a putrid haze that caused scores of university students to choke to death. Most victims died from smoke inhalation rather than burns in what appeared to be the world's deadliest nightclub fire in more than a decade.

Survivors and the police inspector Marcelo Arigony said security guards briefly tried to block people from exiting the club. Brazilian bars routinely make patrons pay their entire tab at the end of the night before they are allowed to leave.

But Arigony said the guards didn't appear to block fleeing patrons for long. "It was chaotic and it doesn't seem to have been done in bad faith because several security guards also died," he told The Associated Press.

Later, firefighters responding to the blaze initially had trouble getting inside the Kiss nightclub because "there was a barrier of bodies blocking the entrance," Guido Pedroso Melo, commander of the city's fire department, told the O Globo newspaper.

Authorities said band members who were on the stage when the fire broke out later talked with police and confirmed they used pyrotechnics during their show.

Police inspector Sandro Meinerz, who coordinated the investigation at the nightclub, said one band member died after escaping because he returned inside the burning building to save his accordion. The other band members escaped alive because they were the first to notice the fire.

"It was terrible inside ? it was like one of those films of the Holocaust, bodies piled atop one another," said Meinerz. "We had to use trucks to remove them. It took about six hours to take the bodies away."

Television images from Santa Maria, a university city of about 260,000 people, showed black smoke billowing out of the Kiss nightclub as shirtless young men who attended the university party joined firefighters using axes and sledgehammers to pound at the hot-pink exterior walls, trying to reach those trapped inside.

Bodies of the dead and injured were strewn in the street and panicked screams filled the air as medics tried to help. There was little to be done; officials said most of those who died were suffocated by smoke within minutes.

Within hours a community gym was a horror scene, with body after body lined up on the floor, partially covered with black plastic as family members identified kin.

Outside the gym police held up personal objects ? a black purse, a blue high-heeled shoe ? as people seeking information on loved ones crowded around, hoping not to recognize anything being shown them.

Teenagers sprinted from the scene after the fire began, desperately seeking help. Others carried injured and burned friends away in their arms. Many of the victims were under 20 years old, including some minors. About half of those killed were men, about half women.

The party was organized by students from several academic departments from the Federal University of Santa Maria. Such organized university parties are common throughout Brazil.

"There was so much smoke and fire, it was complete panic, and it took a long time for people to get out, there were so many dead," survivor Luana Santos Silva told the Globo TV network.

The fire spread so fast inside the packed club that firefighters and ambulances could do little to stop it, Silva said.

Another survivor, Michele Pereira, told the Folha de S. Paulo newspaper that she was near the stage when members of the band lit some sort of flare that started the conflagration.

"The band that was onstage began to use flares and, suddenly, they stopped the show and pointed them upward," she said. "At that point, the ceiling caught fire. It was really weak, but in a matter of seconds it spread."

Guitarist Rodrigo Martins told Radio Gaucha that the band, Gurizada Fandangueira, started playing at 2:15 a.m. "and we had played around five songs when I looked up and noticed the roof was burning."

"It might have happened because of the Sputnik, the machine we use to create a luminous effect with sparks. It's harmless, we never had any trouble with it," he said. "When the fire started, a guard passed us a fire extinguisher, the singer tried to use it but it wasn't working."

He confirmed that accordion player Danilo Jacques, 28, died, while the five other members made it out safely.

Police Maj. Cleberson Braida Bastianello said by telephone that the toll had risen to 233 with the death of a hospitalized victim. He said earlier that the death toll was likely made worse because the nightclub appeared to have just one exit through which patrons could exit.

Officials earlier counted 232 bodies that had been brought for identification to a gymnasium in Santa Maria, which is located at the southern tip of Brazil, near the borders with Argentina and Uruguay.

Federal Health Minister Alexandre Padhilha told a news conference that most of the 117 people treated in hospitals had been poisoned by gases they breathed during the fire. Only a few suffered serious burns, he said.

Brazil President Dilma Rousseff arrived to visit the injured after cutting short her trip to a Latin American-European summit in Chile.

"It is a tragedy for all of us," Rousseff said.

Most of the dead apparently were asphyxiated, according to Dr. Paulo Afonso Beltrame, a professor at the medical school of the Federal University of Santa Maria who went to the city's Caridade Hospital to help victims.

Beltrame said he was told the club had been filled far beyond its capacity.

Survivors, police and firefighters gave the same account of a band member setting the ceiling's soundproofing ablaze, he said.

"Large amounts of toxic smoke quickly filled the room, and I would say that at least 90 percent of the victims died of asphyxiation," Beltrame told the AP.

"The toxic smoke made people lose their sense of direction so they were unable to find their way to the exit. At least 50 bodies were found inside a bathroom. Apparently they confused the bathroom door with the exit door."

In the hospital, the doctor "saw desperate friends and relatives walking and running down the corridors looking for information," he said, calling it "one of the saddest scenes I have ever witnessed."

Rodrigo Moura, identified by the newspaper Diario de Santa Maria as a security guard at the club, said it was at its maximum capacity of between 1,000 and 2,000, and partygoers were pushing and shoving to escape.

Santa Maria Mayor Cezar Schirmer declared a 30-day mourning period, and Tarso Genro, the governor of the southern state of Rio Grande do Sul, said officials were investigating the cause of the disaster.

The blaze was the deadliest in Brazil since at least 1961, when a fire that swept through a circus killed 503 people in Niteroi, Rio de Janeiro.

Sunday's fire also appeared to be the worst at a nightclub since December 2000, when a welding accident reportedly set off a fire at a club in Luoyang, China, killing 309.

In 2004, at least 194 people died in a fire at an overcrowded nightclub in Buenos Aires, Argentina. Seven members of a band were sentenced to prison for starting the flames.

A blaze at the Lame Horse nightclub in Perm, Russia, killed 152 people in December 2009 after an indoor fireworks display ignited a plastic ceiling decorated with branches.

Similar circumstances led to a 2003 nightclub fire that killed 100 people in the United States. Pyrotechnics used as a stage prop by the 1980s rock band Great White set ablaze cheap soundproofing foam on the walls and ceiling of a Rhode Island music venue.

The band performing in Santa Maria, Gurizada Fandangueira, plays a driving mixture of local Brazilian country music styles. Guitarist Martin told Radio Gaucha the musicians are already seeing hostile messages.

"People on the social networks are saying we have to pay for what happened," he said. "I'm afraid there could be retaliation".

VALLECITO?LAKE, Colo. ? Braving cold temperatures and a constant?down pouring of snow, family and friends of 13-year-old Dylan?Redwine came together Saturday to raise attention and awareness for the missing teen.

Redwine, who vanished while visiting his father over Thanksgiving break, has not been seen or heard from in over two months. Friends and private donations have raised over $50,000 towards a reward, but investigators have reported a shortage in tips and leads in recent weeks.

?I think it?s important to keep it out there about the last physical place that Dylan was supposed to be,? Dylan?s mother, Elaine Redwine, said in an interview Saturday near Durango.

More than 25 people showed up outside the missing teen?s father, Mark Redwine?s home, hoping to urge him to be more transparent about his interaction with his son in the hours leading up to Dylan?s disappearance.

Mark Redwine, who investigators have reported as being cooperative with their case, decided to leave the Vallecito Lake area this weekend to avoid confrontation with his ex-wife and others.

?There?s no reason for anybody to be pointing fingers at me,? Mark said in a phone interview with FOX 31 Denver earlier this week. ?Because they want to make me the easiest target to make somebody to blame, I can?t stop them from doing that, but it doesn?t mean I have to agree with it,? he said.

Cory Redwine, the missing teen?s brother, has publically asked his father to be more involved with the search to help bring Dylan back home.

?Right now it?s just a question mark; he was last the person to see him. He hasn?t done his part in helping find Dylan,? Cory explained.

Chile aims to focus summit on its brighter side

SANTIAGO, Chile (AP) ? European, Latin American and Caribbean leaders gathering for this weekend's economic summit will likely see only one side of Chile ? the polished, upscale country where tourists and investors stay in five-star hotels in a sparklingly clean financial district nicknamed "Sanhattan," well away from Santiago's slums.

Hundreds of security agents will ensure that presidents and prime ministers won't be exposed to activists demanding a wider distribution of Chile's copper wealth and decent educations for all. They also won't hear Mapuche Indians denouncing the dictatorship-era anti-terror laws used against Chile's largest indigenous group.

This 16.5 million-person Andean country has won worldwide acclaim for its modernizing economy and institutions, rarities in a region still struggling to leave behind centuries of economic dysfunction. Yet there's another side to the Chilean miracle, one that will sit just blocks from the conference halls and hotels where leaders will meet this week.

Chile, in fact, has the worst inequality rate among the 34 countries of the Organization for Economic Co-operation and Development, which includes other emerging economies such as Mexico and Turkey, according to the World Bank. Its rate of inequality is also worse than several Latin American countries with lower average incomes, including Peru, the Dominican Republic and Ecuador.

"Chile is a country of contrasts. We have two countries, and the people who come to the summit will only see one of them - 'Sanhattan' and the macroeconomic data," said pollster Marta Lagos.

"It might be good for the government that the people in the summits don't realize this, but it's bad for Chile."

Promoting sustainable development by fighting poverty, reducing income inequality and protecting natural resources are actually primary goals of the summit, which brings together more than 60 nations from the European Union, Latin America and the Caribbean, as well as the world's leading non-governmental organizations.

European Commission President Jose Manuel Barroso, Spanish President Mariano Rajoy and German Chancellor Angela Merkel are among those joining a business conference Friday where 300 executives are formally seeking joint ventures with the same goals.

International Monetary Fund Director Christine Lagarde framed their challenges starkly at last week's Davos Conference, saying the world's top executives agree that "severe income disparity" poses the greatest risk to the global economy in the decade ahead. "Excessive inequality is corrosive to growth; it is corrosive to society," she warned.

Other countries, however, can take many positive lessons from Chile.

The world's top copper producer is seen as probably the best managed economy in Latin America because of its strong growth, prudent fiscal and macroeconomic policies and strong institutions that make it an investor's paradise.

But critics say policies launched under former dictator Augusto Pinochet that privatized much of the country are still blocking urgent social reform and fostering social exclusion and inequality.

Pinera's government has been hit by wide protests demanding improvements in education, housing and health care and the protection of the environment from business interests. So while the country is expanding quickly, Chileans are asking for reforms to a system that they say still fails many.

Some of Chile's contradictions are in plain sight. Slum dwellers living in wooden shacks with zinc roofs at Campamento Juan Pablo II have a perfect view of elegant mansions, soaring skyscrapers and luxury car dealerships, just across a wide avenue in Las Condes, one of Santiago's wealthiest neighborhoods.

"The social gap is huge. The millionaires are right there next to us and we're stuck here living in this shantytown. The rich continue to get richer while the poor get poorer," said Raul Sanchez, 51, who earns poverty-level wages watching over cars at nearby parking lots.

No one can deny that President Sebastian Pinera has tried to fight poverty by encouraging job creation and giving cash handouts to the poorest of the poor.

But he has fallen far short of his campaign promise to eradicate poverty by the end of his term in 2014, and Chileans perceive the wealth gap as wider than ever, Lagos said.

Displaying economic success is particularly important to Chile, which is eager to be seen as a developed nation. Chile's business elites take particular pride in the country's membership in the Organization for Economic Cooperation and Development.

The country's economy is expected to grow around 4.8 percent in 2013, outpacing every other country in the region but Peru, according to the United Nations.

Chile's strong standing has helped it weather the world economic crisis and withstand the contagion in the Euro zone, where the economy contracted by 0.5 percent last year. Along with mining, Chile enjoys strong consumer demand and profits from exports of salmon, forestry, wine and fruit. During a recent visit to Santiago, International Monetary Fund Director Christine Lagarde praised Chile for being one of the fastest growing economies in South America and keeping unemployment at historically low levels while controlling inflation below a target 3 percent.

"Chile has been a successful country and there are many reasons why Chileans should be proud, but the country remains largely unequal despite progress," said Patricio Navia, a Chilean political scientist who teaches at New York University.

"Precisely because it has grown so much, one wonders why the government hasn't been able to do more to fight inequality."

Far from the summit's headquarters in a heavily guarded conference center in the Andean foothills, protesters were preparing to take the streets for a "people's summit" on Friday.

Students are demanding an end to the decentralization of education in Chile, which has created a system of failing public schools, expensive private universities, unprepared teachers and pricey student loans.

Mapuche Indians are demanding autonomy and a return of their ancestral territory in Chile's Patagonia region, where timber companies, foreign corporations and wealthy individuals control most of the land. Environmentalists are demanding an end to the privatization of Chile's water, another legacy of the Pinochet era.

The most common complaint of the protesters is that wealth and power has been concentrated in very few hands.

"The biggest problem in Chile is that the differences between the haves and the have-nots are very dramatic," Navia said. "So you are not likely to see poverty as you see in other countries, but you will see that wealth and income in Chile are very unequally distributed."

SANAA, Yemen?? Al-Qaida's No. 2 in Yemen died in a U.S. drone attack last year in southern Yemen, the country's official news agency and a security official said Thursday.

Saeed al-Shihri, a Saudi national who fought in Afghanistan and spent six years in the U.S. military prison at Guantanamo Bay, was wounded in a missile attack in the southern city of Saada on Oct. 28, according to SABA news agency.


The agency said that he had fallen into a coma since then. It was not clear when he actually died.

A security official said that the missile had been fired by a U.S.-operated, unmanned drone aircraft. He spoke on condition of anonymity because he was not authorized to speak to the press.

Yemen had previously announced al-Shihri's death in a Sept. 10 drone attack in the province of Hadramawt. A subsequent DNA test however proved that the body recovered was not that of al-Shihri.

On Oct. 22, al-Shihri denied his own death in audio message posted on Jihadi websites.

Also known by the nom de guerre Abu Sufyan al-Azdi, he denounced at the time the Yemeni government for spreading the "rumor about my death ... as though the killing of the mujahideen (holy warriors) by America is a victory to Islam and Muslims."

Al-Shihri went through Saudi Arabia's famous "rehabilitation" institutes after he returned to his home country, but then he fled to Yemen and became deputy to Nasser al-Wahishi, the leader of an al-Qaida group.

Al-Shihri's death is considered a major blow to al-Qaida's Yemen branch, known as al-Qaida in The Arabian Peninsula. Washington considers it the most dangerous of the group's offshoots.

Al-Qaida in Yemen has been linked to several attempted attacks on U.S. targets, including the foiled Christmas Day 2009 bombing of an airliner over Detroit and explosives-laden parcels intercepted aboard cargo flights last year.

In 2011, a high-profile U.S. drone strike killed U.S.-born Anwar al-Awlaki, who had been linked to the planning and execution of several attacks targeting U.S. and Western interests, including the attempt to down a Detroit-bound airliner in 2009 and the plot to bomb cargo planes in 2010.

Yemen, the Arab world's poorest nation, has fallen into lawlessness since the start of an uprising in 2011, when millions of Yemenis took to the streets demanding the ouster of their longtime authoritarian ruler Ali Abdullah Saleh.

Al-Qaida militants exploited the unrest and took control of large swaths of land in the south until last spring, when the military, backed by the U.S., managed to drive hundreds of militants out of major cities and towns.

Since then, the group has carried out deadly attacks targeting mostly security and military officials, including suicide bombings that targeted military and security compounds.

Gene mutation immortalizes malignant melanoma

Jan. 25, 2013 ? Scientists from the German Cancer Research Center and from University Duisburg-Essen have discovered a previously unknown genetic cause of malignant melanoma: a gene mutation that leads to overactive telomerase, the so-called 'immortality enzyme'. The mutated gene region found in familial melanoma is also altered in up to 74 percent of non-inherited cases of melanoma -- here as a consequence of sun exposure. Substances inhibiting telomerase may be a novel therapeutic approach for treating malignant melanoma. The researchers have published their findings in Science.

About ten percent of all cases of malignant melanoma are familial cases. The genome of affected families tells scientists a lot about how the disease develops. Prof. Dr. Rajiv Kumar of the German Cancer Research Center (Deutsches Krebsforschungszentrum, DKFZ) together with Prof. Dr. Dirk Schadendorf from Essen University Hospital studied a family where 14 family members were affected by malignant melanoma.

The scientists analyzed the genomes of family members and found an identical mutation in the gene for telomerase, an enzyme often called 'immortality enzyme', in all persons studied. Telomerase protects the ends of chromosomes from being lost in the process of cell division and, thus, prevents that the cell ages and dies. The inherited gene mutation leads to the formation of a binding site for protein factors in the controlling region of the telomerase gene, causing it to become overactive. As a result, mutated cells overproduce telomerase and hence become virtually immortal.

This spectacular finding of the family analysis prompted the scientists to also look for mutated telomerase genes in non-inherited (sporadic) melanoma, which is much more common than the familial variant. In most of the tissue samples of melanomas of all stages they found alterations in the telomerase gene switch, which the researchers clearly identified as typical consequences of sun exposure. Even though these mutations were not identical to those found in the melanoma family, they had the same effect: overactive telomerase.

"We don't believe that the telomerase gene in melanoma is mutated by pure chance, but that it is a so-called driver mutation that drives carcinogenesis," says Rajiv Kumar. This is also confirmed by the surprising incidence of this alteration: The telomerase gene is the most frequently mutated gene in melanoma. "This is something we hadn't expected, because malignant melanoma has been genetically analyzed thoroughly. But this mutation always seems to have been overlooked," says Kumar.

Rajiv Kumar, Dirk Schadendorf and their teams are hoping that the alterations in the telomerase gene may be a starting point for developing novel treatment methods for malignant melanoma. A very recent development targeting a specific alteration in the B-RAF gene, which characterizes about half of all melanomas, has shown that this is possible. The mutation gave rise to the development of a targeted drug that can arrest cancer growth. "Substances inhibiting telomerase have already been developed and some of them have even been tested in phase III clinical trials," said Rajiv Kumar. Inhibition of the immortality enzyme might also be able to arrest growth in melanoma.

Shrunken Proton Baffles Scientists

One of the Universe's most common particles has left physicists completely stumped. The proton, a fundamental constituent of the atomic nucleus, seems to be smaller than thought. And despite three years of careful analysis and reanalysis of numerous experiments, nobody can figure out why.

An experiment published today in Science only deepens the mystery, says Ingo Sick, a physicist at the University of Basel in Switzerland. "Many people have tried, but none has been successful at elucidating the discrepancy."

The proton's problems started in 2010, when a paper published in Nature seemed to show that the particle was 4% smaller than originally thought. Researchers began with a target of hydrogen, an atom that consists of one proton and one electron. When they bombarded the hydrogen with muons ? heavier cousins of electrons ? from a particle accelerator, a muon would occasionally replace an electron. Probing the muonic hydrogen with a laser yielded a high-precision measurement of the proton's size. The problem is that the measurement differed from those obtained by two other methods?by 4%, or 0.03 femtometers (fm). That's a tiny amount ? 1 fm is 0.000000000001 millimeter ? but is still significantly larger than the error bars on either of the other measurements.

The latest experiment also used muonic hydrogen, but probed a different set of energy levels in the atom. It yielded the same result as the Nature paper ? a proton radius of 0.84 fm, says Aldo Antognini, a physicist at the Swiss Federal Institute of Technology Zurich in Switzerland and an author of both muonic papers. The second measurement "is totally compatible with the previous value," he says.?

But it is still not compatible with the measurements taken by non-muonic techniques, says John Arrington, a nuclear physicist at Argonne National Laboratory in Lemont, Illinois. Errors in the muon-based measurements of the proton radius are unlikely to be to blame, Arrington says, and yet it seems equally unlikely that all the other measurements are wrong, too.

Perplexing puzzle
One possibility is that Antognini's team has inadvertently discovered new physics. It is the only one to use muons to probe the proton ? the others all used electrons, and there is a small possibility that muons interact with protons differently from electrons. The effect would have to be small, or it would also show up in other places, such as the Large Hadron Collider, the big particle accelerator near Geneva, Switzerland.

Arrington and Sick both have their doubts. "I'm a big believer in our understanding of physics," Arrington says. Given the power of existing theories, Sick says, the idea of fundamental differences between muons and electrons is "sort of hard to imagine".

But equally hard to imagine is what might have gone wrong. Experimentalists have combed back over their data. Theorists have recrunched their equations. There could be a problem with the models used to estimate the proton size from the measurements, but so far, none has been identified. "Many of the ideas that have been stated have all been looked at in more detail," Sick says. "Nobody has come up with a clear result."

Pebble has just informed backers that the very first orders will begin shipping today, though the iOS app is still pending approval from Apple. In the meantime, Android users will have access to a compatible app in Google Play beginning tomorrow -- just in time to start using those freshly unboxed smartwatches. iPhone, iPad and iPod touch users will be able to display notifications and control music in the meantime, though advanced features like watchface and OS updates won't be possible until the app is ready for download.

Now, a bit more bad news: the Pebble team reportedly jumped the gun on shipment notifications, so even though you may have received word that your watch is on the way, it may not be in the first batch of 500. If you've also received a tracking number, however, your device is likely to be in the mail. Scientists have identified genetic circumstances under which common mutations on two genes interact in the presence of cocaine to produce a nearly eight-fold increased risk of death as a result of abusing the drug.

An estimated one in three whites who died of cocaine exposure is a carrier of variants that make cocaine abuse particularly deadly.

The variants are found in two genes that affect how dopamine modulates brain activity. Dopamine is a chemical messenger vital to the regular function of the central nervous system, and cocaine is known to block transporters in the brain from absorbing dopamine after its release.

The same dopamine genes are also targeted by medications for a number of psychiatric disorders. The researchers say that these findings could help determine how patients will respond to certain drugs based on whether they, too, have mutations that interact in ways that affect dopamine flow and signaling.

The scientists had previously identified a total of seven mutations on two dopamine-related genes, some of which were linked to the risk for cocaine abuse death. Years of molecular genetics studies showed that the mutations had specific functions ? a single variant alone was associated with an almost three-fold increase in risk of dying of cocaine abuse ? and led researchers to hypothesize that the variants probably interacted because the genes themselves relied on each other for proper function.

A statistical analysis that dissected the complex interactions among the variants combined with cocaine exposure revealed gene-gene-environment interactions that would dramatically increase the risk of death from cocaine abuse.

"Finding an impact factor of 8 just blew us away," said Wolfgang Sadee, professor of pharmacology and director of the Program in Pharmacogenomics at Ohio State University and senior author of the study. "Beyond that, this represents a new paradigm. Going forward, we can ask whether such interactions do exist between variants that may be a normal variation in the population. These kinds of interactions may underlie the genetics of behavior."

These specific findings apply primarily to whites. The researchers found that a different combination of variants affect the risk of cocaine abuse death in African Americans, and that in this population, some of the variants had protective properties.

The research is published in the online journal Translational Psychiatry.

The mutations are mostly single-nucleotide polymorphisms, or SNPs (pronounced "snips"). Each gene contains two alternative forms ? called alleles ? that are functionally identical in most people. However, in some cases, the activity level, or expression, of an allele can differ from its partner allele in a single gene.

The SNPs described here are on two genes: the dopamine receptor D2, which is a target for antipsychotic drugs, and the dopamine transporter DAT, the main target of cocaine and amphetamines.

The variants' clinical relevance was determined in earlier work led by Sadee that analyzed human brain autopsy tissues of people who had died of cocaine overdose and from age-matched drug-free controls.

The variants identified in this work are harder to detect and analyze than many mutations because these variants have no role in making protein; they exist in deeper and often overlooked regions of genes. Sadee's lab has designed a technique to predict and determine their functions based on measurements of how much messenger RNA, a carrier of genetic information, each specific allele expresses.

Having a defined set of a manageable number of variables then made a statistical analysis both possible and a critical step to more fully understanding the effects of these variants. First author Danielle Sullivan, a doctoral student in biostatistics at Ohio State, built logistic regression models to search for the main effects and interactions among the variants associated with the higher risk of cocaine death.

"A combination of variants turned out to have a high effect on the risk of dying. That is called epistasis ? a gene-gene-environment interaction that is seen only when there is that extra stimulus, in this case the cocaine," Sadee said. "It's a three-way system, which is incredibly complex unless you know beforehand that these variables are all related to each other."

Sadee said consideration of how gene-gene-environment interactions affect the impact of single genes could help solve the mystery of "missing heritability." Scientists know that genes are behind the causes of many diseases and conditions, but to date have been unable to document the complete genetic history of any given disease.

More immediately, what he has discovered about these variants is likely to increase understanding of numerous psychiatric disorders and improve the effectiveness of medical therapies for these problems. Dopamine-related conditions include attention deficit-hyperactivity disorder, bipolar disorder, phobias, anxiety and schizophrenia.

"The gene-gene interaction that we've reported here, eliciting what might be a 'perfect dopamine storm' under cocaine stimulation, could well contribute to other conditions and affect response to drugs such as antipsychotics and amphetamines," Sadee said.

Clinical studies led by his lab so far suggest that gene-gene interactions occurring without an environmental stimulus such as cocaine do appear to help predict response to certain medications.

He is also extending the research to a handful of other genes that affect signaling in the brain.

"Each gene gives us new combinations, each one has novel variants that can be tested in this way. And they may be considered normal variations ? they're not associated with a disease process, but if there are multiple variants together, they may push this whole system in the direction that makes disease more likely or influences individual response to circumstances like stress or drugs," Sadee said.
